Does this mean that all (adult) villains have constant +1 against all PCs?

Sure, but adults are controlled by the GM, who doesn’t roll dice.
No, because villains don’t make rolls.
I think it’s a legit hard move to hand a PC a -1 against a particular villain if, for instance, the PC just failed a Pierce the Mask against the foe. But that’s separate from the Influence mechanic.
Remember though that a Villian with Influence can tell the PC who they really are and have them make the choice to reject that influence or affirm their view of the PC.
