When Monica cash in the debt?

That’s a pretty big Debt to ask for, more than just a small favour.
Regardless, the Debt is spent right away.

Big favor or small favor can change from person to person, i think.
And if her sister exit from home before the month’s end?

You’re right, a favour’s weight is different based on the person. Keep that in mind as you MC, sometimes you can decline a Debt if what is being asked it too big, or you’ll have to make it a Persuade an NPC roll instead with the Debt used to add the +3.
If Monica’s sister reneges on the deal (which she shouldn’t, otherwise you’re cheating as the MC) then Monica should get the Debt back and I would give her an extra one for the trouble.
